TMP/ERG del-TECT Four Color

TMPRSS2/ERG del-TECT™ Four Color FISH Probe is a research-use-only fluorescence in situ hybridization (FISH) assay designed to detect TMPRSS2–ERG gene rearrangements on chromosome 21, including fusion events associated with interstitial microdeletions.  This multi-color probe design enables clear visualization of both classic TMPRSS2–ERG fusion patterns and deletion-driven rearrangements that occur between the two genes. By targeting flanking regions in addition to the fusion partners, the assay provides more confident interpretation of complex signal patterns.

Probe Design & Fluorophores

This four-color probe set targets key regions on chromosome 21q22:

ERG 3’ Region – Red
Spans the 3’ coding region of the ERG gene and participates in fusion detection.

ERG 5’ Region (including HMGN1) – Orange
Covers the upstream portion of ERG, enabling visualization of rearrangement breakpoints.

TMPRSS2 – Green
Targets the TMPRSS2 gene, the androgen-regulated fusion partner of ERG.

DSCAM (Telomeric Control Region) – Aqua
Spans the DSCAM gene and provides distal control context on chromosome 21.

Summary & Scientific Background

The TMPRSS2 and ERG genes are both located on chromosome 21q22. TMPRSS2 encodes a transmembrane serine protease and is regulated by androgen-responsive elements. ERG encodes a transcription factor that belongs to the ETS family and plays a role in gene expression regulation, cellular differentiation, and developmental pathways.
