The NISH Positive Control Probe is an in situ hybridization (ISH) reagent designed to confirm assay sensitivity and verify nucleic acid integrity in FFPE tissue sections. It targets PPIB (Peptidylprolyl Isomerase B, also known as Cyclophilin B) — a constitutively expressed housekeeping gene and widely adopted industry-standard positive control sequence in ISH workflows.
Because PPIB is ubiquitously expressed across human tissue types, a robust signal with this probe confirms that the tissue sample has preserved nucleic acid of sufficient quality and that the ISH assay has been performed correctly. Absence of signal may indicate tissue degradation, processing issues, or protocol failure rather than true target negativity.
