PTEN del-TECT™ Four Color FISH Probe is a research-use-only fluorescence in situ hybridization (FISH) assay designed to detect PTEN gene deletions while helping reduce false signal loss caused by nuclear truncation in FFPE tissue. The probe set combines three closely spaced locus-specific probes with a chromosome 10 centromere control for more confident genomic interpretation.

Probe Design & Fluorophores

This four-color probe set targets regions on chromosome 10 surrounding the PTEN locus:

• PTEN (10q23.31) – Orange
  Primary target probe used to detect PTEN gene deletions.

• WAPAL / BMPR1A (centromeric to PTEN) – Green
  Provides upstream genomic context.

• FAS (telomeric to PTEN) – Aqua
  Provides downstream genomic context.

• Chromosome 10 Centromere (CEP10) – Red
  Used to determine monosomy or polysomy of chromosome 10.

 

Summary & Scientific Background

The PTEN (Phosphatase and Tensin) gene encodes a phosphatase that negatively regulates the PI3K/Akt signaling pathway. PTEN plays a key role in DNA repair, genomic stability, stem cell self-renewal, cellular senescence, and cell migration. Loss of PTEN function is widely studied in cancer and translational research settings.
