Gene Background: NTRK2 is a receptor tyrosine kinase that binds brain-derived growth factor (BDGF) and neurotrophin 4/5 (NT-4/5). The gene is essential to central and peripheral nervous system development and cell survival. NTRK2 rearrangements have been detected in many cancers, including glioblastoma, pilocytic astrocytoma, head and neck squamous cell carcinoma, and lung adenocarcinoma. These rearrangements generate fusion genes encoding hybrid proteins with ligand-independent TK activity, leading to unchecked cell proliferation and tumorigenesis.
